- Peyton's mother, Olivia Manning, was a Homecoming Queen at Mississippi. His father, Archie Manning, was a Mississippi and New Orleans Saints quarterback, and his older brother, Eli, is currently the quarterback for the New York Giants. - At 22 years, five months and 13 days, Peyton was the seventh-youngest starting quarterback in NFL history. Just ahead of him on that same list was his own father (22 years and four months for New Orleans in 1971).

- Shortly after beginning his NFL career, Manning started his own charity, the Peyback Foundation. The Peyback Foundation's mission is to help disadvantaged kids, and focuses its efforts in Louisiana, Tennessee, and Indiana.
- On September 6, 2007, St. Vincent Hospital in Indianapolis renamed its children's hospital to "Peyton Manning Children's Hospital at St. Vincent". Manning and his wife made a donation of an undisclosed amount to St. Vincent's and have had a relationship with the hospital since his arrival in Indianapolis.
